Alleinige Aufgabe des Botanischen Gartens ist die Bewahrung der einheimischen Pflanzenwelt der Kapregion. Es finden sich keine fremden Pflanzen im gesamten Areal. Die Anzahl der zu entdeckenden einheimischen Pflanzenarten wird auf eine 7.000 geschätzt. Unter der typischen Fynbosvegetation befindet sich auch Südafrikas Nationalpflanze, die Protea.

Das Kerngebiet des Botanischen Gartens von Kirstenbosch erstreckt sich über 36 Hektar zu Füssen des Tafelberges auf einer Meereshöhe von 100 m bis knapp 1000 m. Alleine aufgrund dieser Differenz in der Höhenlage ist eine riesige Pflanzenvielfalt garantiert. Das erweiterte Schutzgebiet um den eigentlichen Botanischen Garten erstreckt sich über 600 Hektar.
